GuilbertおよびIscoveは、既知組成の無血清培地にアルブミン、トランスフェリン、レシチン、セレンを補充することにより赤血球やマクロファージの前駆細胞を培養可能であることを明らかにしました。本培地は、ダルベッコ改変イーグル培地(DME)を改変し、セレン、追加のアミノ酸およびビタミン、ピルビン酸ナトリウム、HEPESバッファー、硝酸鉄に代わり硝酸カリウムを含有しています。さらに研究を進めた結果、イスコフ培地は、マウスBリンパ球、骨髄由来造血組織、リポポリサッカリドの刺激を受けたB細胞、Tリンパ球、様々なハイブリッド細胞の増殖をサポートできることが明らかにされました。
