General description

レプトマイシンBは抗真菌性抗生物質であり、CRM1依存性で、NES依存性の核-細胞質移行を阻害する抗腫瘍細胞毒素です(核外移行インヒビター)。 NES含有タンパク質には、HIV-1 REV、アクチン、c-Abl、サイクリンB1、MDM2/p53、IκB、MPF、PKA、MEKなどがあります。

Biochem/physiol Actions

レプトマイシンBは、不飽和分枝鎖脂肪酸で、核外輸送の研究における重要なツールです。 レプトマイシンBは、核外輸送シグナルを含むタンパク質の特異的阻害剤です。 レプトマイシンBは、HIV-1 Revタンパク質およびmRNAのRev-依存性排出などの、分子の核細胞質間移行を阻害します。非常に少量を線維芽細胞に加えると、MEKの核内蓄積が引き起こされます。 レプトマイシンBの影響を受けるその他のタンパク質は、アクチン、c-Abl、サイクリンB1、MDM2/p53、IκB、MPF、およびPKAです。 示唆された阻害機序にはレプトマイシンBのCRM1への直接的な結合が関与しており、この結合が、CRM1調節の保存領域にあるシステイン残基との相互作用を介して、CRM1と核外輸送シグナルを含むタンパク質との結合を遮断するとされています。
