プロテアーゼは、ペプチド結合の加水分解によってタンパク質を分解します。 プロテアーゼは、フッ化フェニルメチルスルホニル（PMSF）やジイソプロピルフルオロホスフェートなどのセリン活性部位阻害剤によって不活化されます。

1ユニットは、特記しない限り、pH 7.5、37°C、1分間で、カゼインを加水分解し、1.0 μmol(181 μg)のチロシンに相当する呈色を生ずる酵素量です(Folin-Ciocalteu試薬で発色)。
