Sigma のペプシンは、パン中の酵素耐性デンプン（RS）の分析において、他の酵素と共に使用されています。また、エンドウ豆やホエイ分離タンパク質の、in vitro胃腸消化のシミュレーションにも使用されています。
ペプシンは、タンパク質の消化に使用されるペプチダーゼであり、抗体からの Fab フラグメントの調製に一般的に使用されます。ブタ胃粘膜由来ペプシンは、マウスの乾燥子宮頚管サンプルの加水分解に使用されています。 製品 P6887 は、凍結乾燥粉末として提供され、食物繊維分析におけるタンパク質の消化に使用されています。

Biochem/physiol Actions

ペプシンは、他の多くのペプチダーゼとは異なり、ペプチド結合のみを加水分解し、アミドまたはエステル結合を分解しません。ペプチド結合のどちらかの側に芳香族酸を持つ、特に、もう一方の残基も芳香族酸またはジカルボキシルアミノ酸であるペプチドを特異的に切断します。芳香族アミノ酸を持つペプチドの近くに硫黄を含むアミノ酸が存在する場合、加水分解の感受性が増加します。ペプシンはまた、フェニルアラニンとロイシンのカルボキシル基側を特異的に切断し、グルタミン酸残基のカルボキシル基側を弱く切断します。バリン、アラニン、またはグリシンの結合は切断しません。Z-L-チロシル-L-フェニルアラニン、Z-L-グルタミル-L-チロシン、または Z-L-メチオニル-L-チロシンは、ペプシン消化の基質として使用されることがあります。ペプシンは、幾つかのフェニルアラニン含有ペプチドによって阻害されます。
選択的切断は、P1位とP1'位にある疎水性残基および芳香族残基に対して起きます。インスリンのβ鎖に含まれる、Phe-Val、Gln-His、Glu-Ala、Ala-Leu、Leu-Tyr、Tyr-Leu、Gly-Phe、Phe-Phe、 Phe-Tyrの各結合を切断します。

Analysis Note

タンパク質量は、E1%/280で測定
至適pHは2～4です。4 M尿素および3 M塩酸グアニジン溶液中で活性を示します。60°Cで安定です。ペプシンはpH8.0～8.5で不可逆的に不活化します。

1ユニットは、pH 2.0、37°C、1分間で、0.001のΔA280を生じる酵素量です(ヘモグロビンを基質とし、TCA可溶性産物として測定)。(最終液量 = 16 mL、光路長 = 1 cm)
