シグマの酵素は、Yarrowia lipolytica由来ユビキノンの抽出前の複合体Iの分解（NADH：ユビキノン酸化還元酵素）に用いられました。Pythium ultimumに対するプロテイナーゼKの作用を調べるために用いられました。また、酵素そのものの自己分解によるアミノ酸の産生を伴わず、沈殿スラリー中で加水分解されるペプチド結合の多様性を最大化するためにも用いられました。
プロテイナーゼKは、DNAやRNAの単離の際にタンパク質分解によるヌクレアーゼの不活性化に有用です。リゾチーム、リボヌクレアーゼAなどの陽イオン性タンパク質に結合したエンドトキシンの除去に使用されます。また、肝臓、酵母、リョクトウのミトコンドリアの単離に有用で、膜上の酵素局在化を評価するために使用されます。さらに、抗原結合部位を露出するためのパラフィン包埋組織切片の処理や、脳組織サンプル中のタンパク質の分解に使用されます。製品P8044は凍結乾燥粉末として供給されます。

Biochem/physiol Actions

プロテイナーゼKは、天然タンパク質に高活性を示します。その特異性は幅広く、未変性状態でも多くの蛋白質を分解します。主にα-アミノ基がブロックされた脂肪族アミノ酸や芳香族アミノ酸のカルボキシ基側のペプチド結合を切断します。至適pHは7.5～9.0で、等電点は8.9です。活性化にはCa2+（1～5 mM）を必要とします。プロテイナーゼKは、ジイソプロピルフルオロリン酸（DFIP）やフッ化フェニルメタンスルホニル（PMSF）に阻害されます。
プロテイナーゼＫは安定した反応性の高いセリンプロテアーゼです。結晶構造および分子構造の研究で得られたエビデンスは、この酵素がスブチリシンファミリーに属し、活性部位が触媒三残基造（Asp39-His69-Ser224）を持つことを示しています。プロテイナーゼＫは広範囲の環境（pH、バッファー塩、界面活性剤［SDS］、温度）で安定です。 0.1～0.5%のSDSの存在下でプロテイナーゼKは活性を保ち、DNA調製において分離されたDNAの完全性を損なうことなくさまざまなタンパク質およびヌクレアーゼを分解します。

1ユニットは、pH 7.5、37°Cで1分間に尿素変性ヘモグロビンを加水分解し1.0 μmolチロシン相当の発色を生ずる酵素量です(Folin-Ciocalteu試薬で発色)。
