Chemical structure: aminoglycoside

Application

胚培養での推奨使用濃度は50 mg/Lです。ストック溶液は、濾過滅菌後、2～8°Cで最大1ヶ月間、-20°Cで長期間保存できます。溶液は37°Cで3日間安定です。開始レベルでの細菌のタンパク質合成を阻害するために使用します。ストレプトマイシン耐性の機序の研究に使用します。細胞培養用途で細菌によるコンタミネーションを阻止するため、ペニシリン等の薬剤と共に用います。

Biochem/physiol Actions

作用機序：ストレプトマイシンは、30SリボソームサブユニットがS12タンパク質に結合することにより原核生物のタンパク質合成を阻害して作用します。その結果、開始複合体から鎖伸長リボソームへの移行を阻害します。 本化合物はミスコードを引き起こすか又は開始を阻害します。

耐性機序：S12リボソームタンパク質の遺伝子であるrpsLの突然変異により、ストレプトマイシンのリボソームへの結合を阻害します。 また、アミノグリコシドホスホトランスフェラーゼはストレプトマイシンを不活性化します。

抗菌スペクトル：ストレプトマイシンはグラム陰性菌及びグラム陽性菌に対して作用します。

Preparation Note

Stock solutions should be prepared with sterile water at concentrations between 20-25 mg/ml.

Disclaimer

Solutions can be stored at 2-8°C for up to a month or at -20°C for more extended periods. At 37°C, solutions are stable for three days.
