Biochem/physiol Actions
SirReal2は、Sirt2、すなわちがん、炎症、神経変性の発生機序に関与しているNAD依存性リジン脱アセチル化酵素の強力な選択的阻害薬です。SirReal2のIC50値は140 nMで、Sirt2に対する選択性は、Sirt1やSirt3の100倍以上です。SirReal2は、リガンド結合に伴う活性部位の構造変化、すなわち活性部位に隣接するポケットに結合してオープン構造内にSirt2を閉じ込めることで作用すると考えられています。SirReal2はHeLa細胞におけるチューブリンの過剰なアセチル化やチェックポイントタンパク質BubR1の不安定化を引き起こしますが、これらの作用はSirt2の阻害と一致しています。
SirReal2は、Sirt2の強力な選択的阻害薬です。
