Application
4つのカルシウムイオンを含む、熱安定性（好熱性）の細胞外メタロエンドペプチダーゼです。コファクターは、亜鉛およびカルシウムです。疎水性アミノ酸残基のN末端側のタンパク質結合を加水分解します。活性の至適pHは8.0、至適温度は70°Cです。pH 5.0～9.5で極めて安定します。サーモリシンは、切断の特異性が低いため、シーケンシングに適した短い断片を生成します。切断の優先性：X-切断-Y-Z、X=アミノ酸；Y=Leu、Phe、Ile、Val、Met、AlaおよびZはPro以外のアミノ酸。Leuに結合したN末端が最も好まれ、次にPheに結合したN末端が好まれます。ペプチドマッピングやタンパク質の構造および立体構造変化の研究のための限定タンパク質分解にしばしば使用されます。

1 unitは、pH 7.5、37°C、1分間で、カゼインを加水分解して1.0 μmol(181 μg)のチロシンに相当する呈色を生じる(Folin-Ciocalteu試薬で発色)酵素量です。
