ダイズ Glycine max （大豆）由来トリプシン阻害薬は、電気泳動の標準分子量マーカーとして使用されています。

Biochem/physiol Actions

このインヒビターはトリプシンに対して作用し、キモトリプシンおよびプラスミンに対してこれよりも弱い作用を及ぼします。 このインヒビターはトリプシン、血漿カリクレイン、凝固因子Ｘと同様のメカニズムをもってプロテアーゼも阻害します。トリプシンインヒビターは、メタロプロテアーゼ、組織ベースのカリクレイン、酸プロテアーゼまたはチオプロテアーゼに対しては作用しません。 このインヒビターは、プロテアーゼ活性部位の1：1の化学量論合成物を生成することで作用し、次にインヒビターの単一のアルギニン-イソロイシン結合が開裂します。 阻害は可逆性でpH依存性です。

ダイズトリプシン阻害薬は2つのジスルフィド結合で架橋した1本のポリペプチドに181個のアミノ酸残基を含む単量体タンパク質です。
トリプシン1単位＝ N-α-ベンゾイル-L-アルギニンエチルエステル（BAEE）を基質として使用するとき、pH 7.6、25℃においてA253が0.001/分

Preparation Note

トリプシンインヒビターは水およびリン酸バッファーに10 mg/mLの濃度で溶けます。 また、平衡塩溶液に1 mg/mLの濃度で溶け、無血清培地にも溶けます。 濃度が10 mg/mLを超える濃縮溶液は濁ることがあり、黄色からアンバーを呈します。 細胞をトリプシン化した後、解離に使用したトリプシン溶液1 mLに対し1 mLのトリプシンインヒビター溶液に1 mg/mLで再懸濁させます。 次にこの細胞懸濁液を1000 rmpで遠心分離し、細胞ペレットを生成します。

溶液は2～8℃で短期間保存したとき活性を保持できます。溶液はアリコートを-20℃で凍結させると安定します。
