The monoclonal is presented at a concentration of 100μg/ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.

Immunohistochemistry: Clone BMK-13 reacts with fresh frozen tissues that were sectioned (6μm), dried for 1 hour at room temperature and fixed in 1:1 solution of methanol and acetone, and airdried for 1 hour. Sections can be stored wrapped in aluminium foil at -70°C until used. For paraffin embedded sections, 4% PFA (3hr) should be used for skin biopsies. 5 μM sections are dewaxed, and rehydrated with water. A proteinase treatment of 0.1% trypsin for 15 minutes in water should be performed prior to blocking and antibody staining.The antibody reacts better in frozen tissues than in paraffin embedded tissues. Western blotting: purified normal human eosinophils were treated with digitonin (5mg/mL) for 10 minutes before boiling in sample buffer and run on a 15% SDS-PAGE. Purified MBP migrates at ~ 14kDa, and typical human samples range from 10-16kDa. ELISA development for MBP estimation.
Optimal working dilutions must be determined by the end user.

CBL419 is specific for eosinophil major basic protein irrespective of the stages of eosinophil activation and can be regarded as a "pan-eosinophil" marker. This antibody will recognize the 23.8 kDa secreated pro-form of MBP (Pregnancy Associated MBP) found in the placenta and serum of pregnant women as well as the 13.8 kDa mature form of the protein which is found in the matrix of the eosinophil large specific granual. It does not cross-react with horse, mouse or rat MBP.
The protein encoded by this gene is the predominant constituent of the crystalline core of the eosinophil granule. High levels of the proform of this protein are also present in placenta and pregnancy serum, where it exists as a complex with several other proteins including pregnancy-associated plasma protein A (PAPPA), angiotensinogen (AGT), and C3dg. This protein may be involved in antiparasitic defense mechanisms as a cytotoxin and helminthotoxin, and in immune hypersensitivity reactions. It is directly implicated in epithelial cell damage, exfoliation, and bronchospasm in allergic diseases.
FUNCTION: SwissProt: P13727 # Cytotoxin and helminthotoxin. Also induces non-cytolytic histamine release from human basophils. Involved in antiparasitic defense mechanisms and immune hypersensitivity reactions. The proform acts as a proteinase inhibitor, reducing the activity of PAPPA. SIZE: 222 amino acids; 25232 Da SUBUNIT: In pregnancy serum, the proform exists as a disulfide- linked 2:2 heterotetramer with PAPPA, as a disulfide-linked 2:2 heterotetramer with AGT, and as a complex (probably a 2:2:2 heterohexamer) with AGT and C3dg. SUBCELLULAR LOCATION: Note=The proform is secreted. The mature protein is found in the matrix of the eosinophil's large specific granule (crystalloid core). TISSUE SPECIFICITY: High levels of the proform in placenta and pregnancy serum; in placenta, localized to X cells of septa and anchoring villi. Lower levels in a variety of other tissues including kidney, myometrium, endometrium, ovaries, breast, prostate, bone marrow and colon.DEVELOPMENTAL STAGE: Levels of the proform increase in serum and placenta during pregnancy. Skin aging is an extremely important medical and social problem in the modern world. Therefore, a goal of the present work was to estimate changes in the numbers of fibroblast-like cells, proliferating cells nuclear antigen-positive cells, CD45-positive cells, mast cells, and eosinophils in human dermis at different ages. Skin specimens from human fetuses that died antenatally from 20 to 40 weeks of pregnancy and humans who died from different causes from 1 day to 85 years of life were used for the study. Results showed a decrease in a total number and the number of proliferating cells nuclear antigen-positive fibroblast-like cells in dermis with progression of age. The numbers of CD45-positive cells and mast cells are gradually increased with aging. Eosinophils are almost absent in dermis independently on age. Mast cells are probably a main factor that potentially can be involved in tissue damage and aging changes in skin. Mast cells should be regarded as an important target for anti-aging therapy.
