      Background InformationLatent-transforming growth factor beta-binding protein 1 (LTBP-1) is also called Transforming growth factor beta-1-binding protein 1 (TGF-beta1-BP-1). A large latent complex of TGFB1 from platelets is composed of the TGFB1 molecule non-covalently associated with a disulfide-bonded complex of a dimer of the N-terminal propeptide of the TGFB1 precursor and LTBP1. LTBP1 does not bind directly to active TGFB1 only to proforms. LTBP1 appears to be involved in the secretion of TGFB1 to sites at which it is stored or activated, and it is critical in controlling the action of TGFB1. LTBP1 may also play a role in extracellular matrix building and calcium binding mediated signaling surrounding TGFB1 activated signaling pathways. Also, because of its critical role in TGFB1 metabolism, LTBP1 is an important player in many developmental processes including embryo implantation and growth. LTBP1 is also a player in at least two distinct breast cancer metastasis signatures and has both a long and short isoform that work to establish and maintain the spatial restriction of TGFbeta bioavailability during development of many tissues including mammary development.
