      Background InformationMeprin A subunit beta (EC 3.4.24.63; UniProt Q16820; also known as Endopeptidase-2, Meprin B, N-benzoyl-L-tyrosyl-P-amino-benzoic acid hydrolase subunit beta, PABA peptide hydrolase, PPH beta) is encoded by the MEP1B gene (Gene ID 4225) in human. Meprins are multi-domain zinc-containing proteinases that are members of the astacin family and metzincin superfamily of metalloproteinases. Meprins are expressed in epithelial cells in the kidney and intestine, as well as in skin, leukocytes, and lung. The meprin α and β subunits differ in their substrate specificities. Meprin α has a preference for small or hydrophobic residues flanking the cleavage site, whereas meprin β has a preference for negatively charged residues. Both subunits are synthesized as type 1 membrane spanning proteins, however, the transmembrane and cytoplasmic portion of meprin α, but not β, is proteolytically cleaved off in the endoplasmic reticulum. The meprin subunits form disulfide-linked homo- and heterodimers. Meprin β dimers (called meprin B) and meprin α/β dimers (heteromeric meprin A) are membrane-bound proteins, whereas meprin α homodimers are secreted and tend to self-associate into larger oligomers of over 10 subunits.
