ACCUSPIN™チューブは、高密度なポリエチレンバリアーを持った、ポリプロピレン放射滅菌チューブです。各チューブは、15 mLの密度勾配が可能です。
Application
ACCUSPIN™ Tubes Sterile, 50 mL Capacityは、以下の用途で、濃度勾配分離培地であるHistopaque™-1077とともに使用することを意図しています：
- リンパ球およびその他の単核細胞の分離。
- 臍帯血由来内皮前駆細胞の分離。
- 骨髄検体からの単核細胞層の分離。
- 急性骨髄性白血病に対するBET阻害薬の治療効果を検討するためのヒト骨髄吸引液の精製に適しています。
Biochem/physiol Actions
ACCUSPINチューブにHistopaque-1077を追加してから短時間遠心分離すると、Histopaque-1077はフリットの下に配置されます。チューブの上部チャンバーに血液サンプルを追加できるため、フリットの下の下部チャンバー内のHistopaque-1077と混ざるおそれはありません。その直後の遠心分離では、全血がフリットを通って下方に流れてHistopaque-1077と接触します。より密度の高い成分は、フリットの上のHistopaque-1077の容量を変化させて、血液成分が明確に分離されます。赤血球は凝集し、顆粒球はやや高張して沈降速度が速まり、その結果ACCUSPINチューブの底でペレット化が生じます。リンパ球と、単球などのその他の単核球は、血漿／Histopaque-1077界面にとどまります。この単核球の密度帯は、上部チャンバーの中身を注ぎ出すか、またはピペットを用いて採取されます。赤血球汚染は、チャンバー間にバリアがあるため生じません。
