General description

リポ多糖（LPS）は、脂質と多糖で構成される複雑な分子であり、主にグラム陰性菌の外壁に存在します。脂質A、コアオリゴ糖（コア）、およびO-特異的多糖またはO抗原（OAg）で構成されるLPSは、細菌細胞外膜に固定され、脂質Aが主としてその関連毒性に関与します。炎症反応の強力な活性化因子として機能するLPSは、マクロファージ、単球、および内皮細胞を含むさまざまな哺乳動物細胞と結合します。LPSは、グラム陰性菌を環境ストレスから保護する役割を果たすとともに、感染中に自然免疫系による認識をトリガします。

Application

このLPSセロタイプは、B細胞の刺激や、ヒト肝細胞におけるNOSの誘導に使用されています。
リポ多糖（LPS）は、グラム陰性菌の細胞壁に特徴的な成分です。LPSとその成分である脂質Aは、一般的な病原体関連分子パターン（PAMP）を認識するトール様受容体タンパク質ファミリーの1つである、トール様受容体4（TLR4）による先天的な免疫システムの細胞を刺激します。

Biochem/physiol Actions

LPSは、ほとんどのグラム陰性菌の細胞壁を構成する主要成分です。LPSは、可溶性の抗原に対する免疫応答を亢進する能力のある免疫原性の高い抗原です。LPSは、マウス、ウサギ、ニワトリ、雌ウシ、ハムスター、ヒトの骨髄由来Bリンパ球の特異的マイトジェンとしても作用します。

Preparation Note

リポポリサッカリドは、γ線照射処理を行った凍結乾燥粉末です。再構成するには、バイアル(1 mg)に滅菌平衡塩類溶液または組織培養液1 mLを添加し、粉末が溶解するまで穏やかに攪拌します。本製品を再構成後、滅菌平衡塩類溶液または組織培養液を使用して、望ましい使用濃度に希釈できます。
