General description

RNアーゼA、すなわちリボヌクレアーゼAは、ピリミジンヌクレオチドの後でホスホジエステル結合の一本鎖RNAを切断するエンドリボヌクレアーゼです。RNアーゼAは3′リン酸末端を攻撃します（たとえば、pG-pG-pC-pA-pGが切断されて、pG-pG-pCpとA-pGになります)。一本鎖RNAに対して最も高い活性を示します。RNアーゼAは、4箇所でジスルフィド架橋された一本鎖ポリペプチドです。RNアーゼBとは異なり、糖タンパク質ではありません。DNAは環状中間体の形成に必要な2′-OH基を持たないため、リボヌクレアーゼはDNAを加水分解しません。また、RNアーゼAは、タンパク質サンプル中のRNAを加水分解できます。RNアーゼAは、His12とHis119のアルキル化によって阻害され、カリウム塩およびナトリウム塩によって活性化されます。RNアーゼは、重金属イオン存在下で阻害されます。また、RNアーゼは、DNAによって競争的に阻害されます。
