      Application CommentsExhibits staining of the connective tissue matrix when used in immunohistochemistry. This antibody is suitable for both reduced and non-reduced protein by immunoblotting. For staining of formalin-fixed tissues, boil in 10 mM citrate buffer, pH 6.0, for 10-20 min, followed by cooling at room temperature for 20 min. This antibody has also been reported to work for immunofluorescence. Antibody should be titrated for optimal results in individual systems.

      DescriptionProtein G purified mouse monoclonal antibody. Recognizes the ~440 kDa disulfide-linked, dimeric fibronectin.
      BackgroundFibronectin is a dimeric, disulfide-linked glycoprotein of ~440 kDa that is present in basement membranes, extracellular matrix, and blood. Fibronectin contains multiple sites for various ligands and cells including binding sites for collagen, glycosaminoglycans, transglutaminase, and a cell surface receptor. This glycoprotein is involved in cell adhesion, tissue organization, and wound healing. The region of fibronectin that mediates cell adhesion in a majority of cell types is in the C-terminal domain and involves the binding of the Arg-Gly-Asp (RGD) sequence to a specific polypeptide cell surface receptor. Although the mechanisms of fibronectin extracellular matrix assembly are not well understood, specific antibodies have been used to determine the domains involved in matrix assembly. Cellular fibronectin is widely distributed in the stroma of many malignant tumors.
