      Applications
      Key Applications Immunoblotting (Western Blotting)
      Application NotesImmunoblotting (1:1000-1:10,000)
      Immunoprecipitation (1-4 µg/mg lysate)
Application CommentsHAUSP protein is reported to stabilize p53 through the deubiquitination of p53. HAUSP has also been reported to be cleaved by caspase-3. May recognize additional bands, possibly corresponding to HAUSP cleavage products. Antibody should be titrated for optimal results in individual systems.

DescriptionImmunoaffinity purified rabbit polyclonal antibody. Recognizes the ~135 kDa HAUSP protein.
Backgroundp53, a tumor suppressor phosphoprotein is present at low concentrations in normal cells. The low concentration is maintained by MDM2-mediated ubiquitination and resulting proteolysis. HAUSP, an ubiquitin hydrolase, is thought to regulate MDM2 and help stabilize p53. When cells are subjected to stress, such as hypoxia or UV radiation, p53 is activated, its protein levels are elevated and its ubiquitin-dependent degradation is blocked. The resulting increase in p53 leads either to the induction of cell cycle arrest or apoptosis.
