Lanes 1, 2: Detection of human PARC/H7-AP1 by immunoblotting (Lanes 1 and 2). Samples: Whole cell lysate (40 µg) from untreated U2OS cells (Lane 1) or U2OS cells that stably express a short hairpin RNA (shPARC) against PARC/H7-AP1 (Lane 2). Primary antibody: Anti-PARC/H7-AP1 (900-950) Rabbit pAb (Cat. No. DR1028) (1:2500). Detection: chemiluminescence. Lanes 3, 4: Detection of human PARC/H7-AP1 by immunoprecipitation followed by immunoblotting. Samples: Whole cell lysate (500 µg) from untreated U2OS cells. Antibodies for IP: Normal rabbit IgG (2 µg/mg lysate) (Lane 3) and Anti-PARC/H7-AP1 (Cat. No. DR1028) (2 µg/mg lysate) (Lane 4). Immunoblotting conditions: same as Lanes 1 and 2.

DescriptionImmunoaffinity purified rabbit polyclonal antibody. Recognizes the ~270 kDa PARC/H7-AP1 protein.
Backgroundp53 is a tumor suppressor that can induce cell growth arrest, apoptosis, and cellular senescence in response to various types of cellular stress. Tight regulation of p53 is necessary for maintaining normal growth and tumor suppression. In unstressed cells wild-type p53 is a short-lived protein found at low levels. One mechanism for the regulation of p53 is compartmentalization. Upon exposure to DNA damaging agents or other cellular stress p53 translocates from the cytoplasm into the nucleus where it activates endogenous targets. PARC/H7-AP1 (Locus Link ID: 23113)is a Parkin-like ubiquitin ligase that serves as a cytoplasmic anchor for p53-associated protein complexes. PARC has been reported to form a large complex with p53 in the cytoplasm of unstressed cells. Inactivation of PARC/H7-AP1 induces p53 nuclear localization and apoptosis.
