      Background InformationCD81 antigen (UniProt: P35762; also known as 26 kDa cell surface protein TAPA-1, Target of the antiproliferative antibody 1, CD81) is encoded by the Cd81 (also known as Tapa1) gene (Gene ID: 12520) in murine species. TAPA1 is a non-glycosylated, homodimeric, multi-pass membrane protein of the tetraspanin (TM4SF) family. It is expressed in a wide range of cells, including hematolymphoid, neuroectodermal, and mesenchymal tumor cell lines and is reported to play a role in the regulation of lymphoma cell growth. It contains four transmembrane domains, two extracellular loops, and short amino and carboxyl termini that are intracellular. TAPA1 is a structural component of specialized membrane microdomains known as tetraspanin-enriched microdomains (TERMs) that act as platforms for receptor clustering and signaling. It is shown to affect cell adhesion, morphology, activation, proliferation, and differentiation of B, T, and other cells. It is essential for trafficking and compartmentalization of CD19 receptor on the cell surface of activated B cells. Upon initial encounter with a microbial pathogen, it enables the assembly of CD19-CR2 and B cell receptor complexes at signaling TERMs, lowering the threshold dose of antigen required to trigger B cell clonal expansion and humoral immune response. In T cells, it associates with CD4 or CD8 co-receptors and defines the maturation state of antigen-induced synapses with B cells.
