Recognizes the ~130 kDa IFNAR1 protein phosphorylated at Ser535,539.
      Catalogue NumberPS1003
      Brand Family Calbiochem®
      Application Data
Detection of human IFNAR1, phosphorylated at Ser535/539, by immunoblotting. Samples: Whole cell lysate from HEK293 cells mock transfected (lane 1), transfected with wild-type IFNAR1 (lane 2), or transfected with S535A/S539A mutant IFNAR1 (lane 3). Primary antibody: PhosphoDetect™ Anti-IFNAR1 (pSer535,539) Rabbit pAb (Cat. No. PS1003) (1:1000) pre-incubated without peptide (control, left panel), with immunogen phosphopeptide (phos-peptide, middle panel), or non-phosphopeptide corresponding to the phosphopeptide immunogen (dephos-peptide, right panel). Detection: chemiluminescence.

DescriptionRabbit polyclonal antibody adsorbed against non-phosphopeptide corresponding to the immunogen, followed by immunoaffinity chromatography. Recognizes the IFNAR1 protein phosphorylated at Ser535/539.
BackgroundInterferons (IFNs) are important therapeutic agents. They have demonstrated activity in infectious autoimmune diseases and selected cancers. IFNs activate their target cells by first binding to specific receptors on the cell surface to induce synthesis of effector protein. There are three types of natural IFNs: alpha (α), beta (β), and gamma (γ). The first two are classified as type I because they bind to IFN cell surface receptor type I (IFNAR1). IFN γ belongs to type II interferons since it binds to type II receptors only. The stimulation of type I and type II IFNRs results in the activation of the Janus family protein tyrosine kinases (PTKs), i.e. Jak1 and Tyk2 PTKs for IFNAR1, and Jak1 and Jak2 PTKs for type II IFNR. Down-regulation of IFNAR1 plays a role in defining the magnitude and duration of cytokine signaling. This down-regulation is believed to be regulated, in part, by phosphorylation at Ser535 and Ser539.
