      BackgroundEukaryotic cells arrest growth at discrete stages in the cell cycle called checkpoints in order to repair/replace damaged DNA. Checkpoints are also essential for orderly progression and completion of DNA replication and chromosome segregation. Failure to execute a checkpoint can lead to genomic instability and subsequent tumor development. All eukaryotic organisms contain multiple genes required for executing DNA damage checkpoints. The human ATR/FRP1 gene, initially identified on the basis of its sequence similarity to the human FRAP/mTOR, S. cerevisiae MEC1/ESR, S. pombe Rad3, and D. melanogaster Mei-41 genes, encodes a serine/threonine kinase involved in DNA damage response pathways. ATR is also member of a growing family of phosphatidylinositol kinase-related (PIK-R) genes, characterized by their large size and encoding proteins greater than 200 kDa; additional PIK-R genes include the ATM gene (Ataxia-Telangiectasia Mutated) and the catalytic subunit of DNA-PK (DNA dependent protein kinase). Introduction of human ATR into mec1 mutants S. cerevisiae corrects some but not all of the mec1 defects. ATR may also be critical for meiotic checkpoints since the protein binds unsynapsed chromosomes. Recent work confirms a role for ATR in the DNA damage G2/M checkpoint since human cells over-expressing a mutant ATR exhibit increased sensitivity to DNA damaging agents and are unable to arrest at the G2/M checkpoint. The PIK-R kinases can be inhibited by wortmannin, a known radiosensitizer, although ATR is less sensitive to Wortmannin (Cat. No. 681675) than the ATM and DNA-PK kinases.
