      Background InformationCoagulation factor XII (UniProt: P00748; also known as EC: 3.4.21.38, Hageman factor, HAF) is encoded by the F12 gene (Gene ID: 2161) in human. Factor XII is a serum glycoprotein that participates in the initiation of blood coagulation, fibrinolysis, and the generation of bradykinin and angiotensin. It activates factor XI to XIa and factor VII to VIIa by selective cleavage of Arginine-Isoleucine bond. Factor XII itself can be activated to factor XIIa either by exposure to negatively charged surfaces or via proteolytic cleavage. It is synthesized in the liver with a signal peptide (aa 1-19), which is subsequently cleaved off in the mature form. Coagulation factor XII circulates in blood as a single chain zymogen, which is converted to a two-chain serine protease with a heavy chain (alpha-factor XIIa) and a light chain. The heavy chain contains two fibronectin-type domains, two epidermal growth factor (EGF)-like domains, a kringle domain, and a proline-rich domain. The light chain contains only a catalytic domain. Mutations in F12 gene can lead to Factor XII deficiency that is generally an asymptomatic anomaly of in vitro blood coagulation. However, some mutations may cause hereditary angioedema 3, which is characterized by episodic local swelling involving subcutaneous or submucous tissue of the upper respiratory and gastrointestinal tracts, face, extremities, and genitalia.
