      Background InformationGuanine nucleotide-binding protein G(i) subunit alpha-1 (UniProt: P63096; also known as Adenylate cyclase-inhibiting G alpha protein, Gia-1) is encoded by the GNAI1 gene (Gene ID: 2770) in human. The heterotrimeric G-proteins mediate signaling from a large number of seven transmembrane spanning cell surface receptors, commonly known as G-protein coupled receptors (GPCRs), to a variety of intracellular effectors. These signal transduction pathways control numerous essential functions in all tissues and are ubiquitous throughout the animal kingdom. The binding of GTP produces a conformational change in the G-protein a-subunit resulting in the dissociation of the receptor-G-protein complex and freeing the and bg subunits to interact with their target effectors. The slow intrinsic GTPase activity of the subunit hydrolyzes the bound GTP to GDP ending the interaction of the a-subunit with its effector. G-proteins are composed of a 36-52 kDa a subunit, a 35-36 kDa b-subunit and an 8-10 kDa g-subunit. G-proteins are classified by the nature of their a-subunit, which are grouped into four general classes (as, ai/o, aq and a12) based on amino acid similarities. Most of the subunits are widely distributed across tissue types except for the subunits mediating sensory transduction. Each subclass of G-protein targets a specific signaling cascade. Ga(s) are known to regulate adenylate cyclase where Ga(s) stimulates adenylate cyclase activity, and Ga(i) and Ga(o) perform an inhibitory role. The functional properties of the subunits are, in part, determined by the covalent attachment of various lipids. Reversible palmitoylation of N-terminal cysteines has been described for all subunits except transducin, while the non-reversible myristoylation of an N-terminal glycine is restricted to members of the i/o family. These modifications facilitate membrane association and functional interactions with other components of the signal transduction systems, especially the bg subunits and certain effector molecules.
