      Background InformationReceptor-type tyrosine-protein phosphatase delta (UniProt: Q64487; also known as EC:3.1.3.48, Protein-tyrosine phosphatase delta, R-PTP-delta) is encoded by the Ptprd gene (Gene ID: 19266) in murine species. R-PTP-delta is a single-pass type I membrane protein that is expressed in various regions of the brain, and in kidney, heart and some B-cell lines. It is one of the receptor type IIa protein tyrosine phosphate that plays a role in axon guidance, synapse formation, cell adhesion, and tumor suppression. It is shown to bidirectionally induce pre- and post-synaptic differentiation of neurons by mediating interaction with IL1RAP and IL1RAPL1 trans-synaptically and is also involved in pre-synaptic differentiation through interaction with SLITRK2. Mouse brain is shown to express two major isoforms of R-PTP-delta corresponding to C and D isoforms with molecular weights of 130 and 80 kDa, respectively. Its expression increases from P11 and reaches a maxim level at P23. R-PTP-delta knockout mice are reported to be semi-lethal and display signs of motor dysfunction. R-PTP-delta is synthesized with a signal peptide (aa 1-20), which is subsequently cleaved off to generate the mature form that contains a large extracellular domain (aa 21-1266), a transmembrane domain (aa 1267-1287), and a cytoplasmic domain (aa 1288-1912). It has three Ig-like C2-type domains in its N-terminal region, eight Fibronectin type-III domains, and two tyrosine-protein phosphatase 2 domains in the C-terminal region (aa 1357-1612 and 1644-1903). It can undergo a cleavage between residues 1181-1182 that separates the extracellular domain from the transmembrane segment.
