      ApplicationImmunoblotting (1 µg/ml, chemiluminescence)
Paraffin Sections (1 µg/ml, heat pre-treatment required; see application references)
DescriptionPurified mouse monoclonal antibody generated by immunizing mice with the specified immunogen and fusing splenocytes with Sp2/0 mouse myeloma cells. Recognizes the ~27 kDa p27kip-1 protein.
Backgroundp27, the product of the p27Kip1 gene, is an inducible inhibitor of cyclin dependent kinase activity. Related to p21WAF, p27 interacts strongly with D-type cyclins in complex with cdk4 and more weakly with cyclin E and cdk2 complexes2. Based on results from experiments in which p27 is overexpressed in cell lines, it appears that p27 acts as a negative regulator of G1 progression, and in fact, increased levels of the protein have been observed in cells undergoing G1 arrest following treatment with TGFβ. The p27 gene has been localized to chromosome 12p13.
